Indication:
This medication is used to treat ovarian cancer. It is a chemotherapy drug that works by slowing the growth of cancer cells. It may also be used for other conditions as determined by your doctor. Lynparza (Olaparib) is a PARP enzyme inhibitor. It works by killing certain cancer cells.
Mechanism of action:
Lynparza (Olaparib) is an inhibitor of poly (ADP-ribose) polymerase (PARP) enzymes, including PARP1, PARP2, and PARP3. PARP enzymes are involved in normal cellular homeostasis, such as DNA transcription, cell cycle regulation, and DNA repair. Lynparza (Olaparib) has been shown to inhibit growth of select tumor cell lines in vitro and decrease tumor growth in mouse xenograft models of human cancer both as monotherapy or following platinum-based chemotherapy. Increased cytotoxicity and anti-tumor activity following treatment with Lynparza (Olaparib) were noted in cell lines and mouse tumor models with deficiencies in BRCA. In vitro studies have shown that Lynparza (Olaparib)-induced cytotoxicity may involve inhibition of PARP enzymatic activity and increased formation of PARP-DNA complex, resulting in disruption of cellular homeostasis and cell death.
Dosage and Administration:
- Lynparza (Olaparib) is a pill, taken by mouth, twice daily. It can be taken with or without food.
- Take Lynparza (Olaparib) exactly as prescribed.
- Swallow Lynparza (Olaparib) capsules whole. Do not crush, dissolve or open capsules.
- Do not take Lynparza (Olaparib) capsules if they look damaged or show signs of leakage.
- Do not change your dose or stop Lynparza (Olaparib) unless your health care provider tells you to.
- If you miss a dose, take your next dose at your usual scheduled time. Do not take an extra dose to make up for a missed dose.
- If you take too much Lynparza (Olaparib), call your health care provider right away and go to the emergency room.
- Avoid grapefruit juice and Seville oranges during treatment with Lynparza (Olaparib). Grapefruit and Seville oranges may increase the level of Lynparza (Olaparib) in your blood.
Side effects:
Nausea, vomiting, loss of appetite, diarrhea, constipation, bad taste in your mouth, dizziness, or joint/back/muscle pain may occur. Rarely, nausea and vomiting can be severe. In some cases, your doctor may prescribe medication to prevent or relieve nausea and vomiting. Eating several small meals or limiting activity may help lessen nausea and vomiting. If any of these effects persist or worsen, tell your doctor or pharmacist promptly.
Precaution:
Before starting Lynparza (Olaparib) treatment, make sure you tell your doctor about any other medications you are taking (including prescription, over-the-counter, vitamins, herbal remedies, etc.).
Do not receive any kind of immunization or vaccination without your doctor's approval while taking Lynparza (Olaparib).
Store: Store Lynparza (Olaparib) at room temperature, between 68 and 77 degrees F (20 and 25 degrees C). Store away from heat, moisture, and light. Avoid temperatures higher than 104 degrees F (40 degrees C). Do not take Lynparza (Olaparib) if it has been at a temperature higher than 104 degrees F (40 degrees C). Keep Lynparza (Olaparib) out of the reach of children and away from pets.
